中文摘要: 目的:为木鳖子良种选育与高产优质栽培提供参考。方法:在2015年版《中国药典》(一部)等文献基础上,优化测定木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量的高效液相色谱(HPLC)法;比较8个省14个产地的木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量并进行聚类分析;采用SPSS 17.0软件分析海拔、经纬度与木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量间的相关性。结果:木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯质量浓度在0.05~0.5 mg/mL范围内与峰面积呈良好线性关系,回归方程为Y=4 361.95X+67.380 8(R2=0.999 7);定量限为15.62 ng,检测限为4.67 ng;平均回收率为99.76%(RSD=1.36%,n=9)。8个省14个产地的木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量存在极显著差异(P<0.01)。根据聚类分析结果,将8个省14个产地的木鳖子划分为Ⅰ、Ⅱ、Ⅲ、Ⅳ4个类群,Ⅲ与Ⅳ类群木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量较高,包括广西壮族自治区平南县、贵州省江口县、贵州省德江县、福建省建阳区与湖南省会同县5个产地,其中广西壮族自治区平南县、贵州省江口县、贵州省德江县、湖南省会同县属于高海拔、低经纬度地区,福建省建阳区属于高海拔地区。木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量与海拔呈正相关,与经度、纬度呈负相关。结论:测定木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量的HPLC法简便、准确、重复性好,所测8个省14个产地的木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯含量均达到2015年版《中国药典》(一部)标准,广西壮族自治区平南县、贵州省江口县、贵州省德江县、湖南省会同县等低经纬度、高海拔地区栽培有利于积累木鳖子中丝石竹皂苷元3-O-β-D-葡糖醛酸甲酯成分。
英文摘要: OBJECTIVE: To provide reference for variety breeding, high yield and high quality cultivation of Momordica cochinchinensis. METHODS: Based on Chinese Pharmacopoeia (2015 edition, Vol Ⅰ), HPLC for determining the content of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis was optimized, the contents of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis from 14 production places of 8 provinces were compared, and cluster analysis was conducted. Correlation of longitude, latitude and altitude with content of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis was analyzed by SPSS17.0 software. RESULTS: Gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis showed good linear relationship in 0.05-0.5 mg/mL, regression equation was Y=4 361.95X+67.380 8(R2=0.999 7); the limits of quantification and detection were 15.62 ng and 4.67 ng, respectively. Average recovery was 99.76% (RSD=1.36%, n=9). The content of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis from 14 production places of 8 provinces had extremely significant differences (P<0.01). Clustering analysis results indicated that M. cochinchinensis from 14 production places of 8 provinces were divided into Ⅰ, Ⅱ, Ⅲ, Ⅳ groups. Contents of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis of Ⅲ, Ⅳ groups were relatively high, including Guangxi Pingnan, Guizhou Jiangkou, Guizhou Dejiang, Fujian Jianyang and Hunan Huitong, in which, Guangxi Pingnan, Guizhou Jiangkou, Guizhou Dejiang and Hunan Huitong had high altitude and low latitude, Fujian Jianyang had high altitude. The content of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis was positively correlated with altitude, while negatively correlated with longitude and latitude. CONCLUSIONS: HPLC is simple, accurate and reproducible for determining the content of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis, and the determined contents of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis from 14 production places of 8 provinces have reached the standard of Chinese Pharmacopoeia (2015 edition, Vol Ⅰ). Cultivation in Guangxi Pingnan, Guizhou Jiangkou, Guizhou Dejiang, Hunan Huitong and other areas with high altitude and low latitude helps to improve the content accumulation of gypsogenin 3-O-β-D-glucuronic acid methyl ester in M. cochinchinensis.
