中文摘要: 目的:筛选太子参抗心肌细胞缺氧/复氧(H/R)损伤的活性部位,并探讨其作用机制。方法:太子参药材水提物经D101大孔吸附树脂柱以水-乙醇梯度洗脱,得水洗脱部位(Fr.A)、30%乙醇洗脱部位(Fr.B)、50%乙醇洗脱部位(Fr.C)、95%乙醇洗脱部位(Fr.D)。采用化学缺氧法以连二亚硫酸钠复制大鼠H9c2心肌细胞H/R损伤模型,采用MTS法检测经太子参各部位低、中、高剂量(Fr.A、Fr.B、Fr.C低、中、高剂量均分别为750、1 500、3 000 mg/L,Fr.D低、中、高剂量分别为5、10、20 mg/L)预处理后的细胞存活率,筛选活性最强部位。分别采用微板法、硫代巴比妥酸比色法、WST-1法测定经太子参活性最强部位低、中、高剂量预处理后细胞培养液中乳酸脱氢酶(LDH)活性以及细胞内丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性,采用流式细胞术检测细胞凋亡率,采用蛋白质印迹法检测细胞胱天蛋白酶3(Caspase-3)、B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)的表达水平。结果:经4个部位预处理后,Fr.A高剂量组,Fr.B高剂量组,Fr.C各剂量组的细胞存活率均较模型组显著升高(P<0.05或P<0.01),且以Fr.C的活性最强。经Fr.C预处理后,各剂量组细胞培养液中LDH活性,中、高剂量组细胞内MDA含量、细胞凋亡率及Caspase-3和Bax蛋白的表达量均较模型组显著下降(P<0.05或P<0.01);高剂量组细胞内SOD活性以及中、高剂量组细胞Bcl-2蛋白的表达量均较模型组显著升高(P<0.05)。结论:太子参Fr.A、Fr.B、Fr.C部位均具有抗心肌细胞H/R损伤的作用,其中以Fr.C的活性最强;Fr.C抗心肌细胞H/R损伤作用可能与其改善细胞的氧化应激状态,提高细胞清除氧自由基的能力,降低细胞脂质过氧化程度,抑制细胞凋亡,上调Bcl-2蛋白表达以及下调Caspase-3、Bax蛋白表达有关。
英文摘要: OBJECTIVE: To screen the active fractions of Pseudostellaria heterophylla against myocardial hypoxia/reoxygenation (H/R) injury, and to investigate its mechanism. METHODS: The water extract of P. heterophylla was eluted with D101 macroporous adsorption resin column by water-ethanol gradient elution to obtain water elution fraction (Fr.A), 30% ethanol eluting fraction (Fr.B), 50% ethanol eluting fraction (Fr.C) and 95% ethanol eluting fraction (Fr.D). H/R injury model of rat H9c2 myocardial cells was induced by chemical anoxia method with natrium hydrosulfurosum. MTS assay was used to detect survival rate of cells after pretreated with low-dose, medium-dose and high-dose of each fraction (low-dose, medium-dose and high- dose of Fr.A, Fr.B, Fr.C were 750, 1 500, 3 000 mg/L;low-dose, medium-dose and high-dose of Fr.D were 5, 10, 20 mg/L), and the strongest active fraction of P. heterophylla was screened. After pretreated with the strongest active fraction, the activity of LDH in cell culture medium, the content of MDA and the activity of SOD in the cells were determined by microplate method, TBA method and WST-1 method, respectively. The apoptotic rate of the cells was detected by flow cytometry. The expressions of Caspase-3, Bcl-2 and Bax were detected by Western blot. RESULTS: After the pretreatment of 4 fractions, the survival rate of cells in Fr.A high-dose group, Fr.B high-dose group and all Fr.C groups were increased significantly, compared with model group (P<0.05 or P<0.01). The activity of Fr.C was the strongest. After pretreated with different doses of Fr.C, the activity of LDH in the culture medium of all groups, the content of MAD, apoptotic rate, the protein expression of Caspase-3 and Bax in medium-dose and high-dose groups were all decreased significantly, compared with model group (P<0.05 or P<0.01); the activity of SOD in high-dose group, the protein expression of Bcl-2 in medium-dose and high-dose groups were increased significantly, compared with model group (P<0.05). CONCLUSIONS: The Fr.A, Fr.B and Fr.C of P. heterophylla have anti-H/R injury effects on cardiomyocytes, and the activity of Fr.C is the strongest. Anti-H/R injury of Fr.C may be related to its ability to improve oxidant stress state and scavenge oxygen free radicals, decrease the degree of lipid peroxidation, inhibit cell apoptosis, up-regulate the protein expression of Bcl-2 and down-regulate the protein expression of Caspase-3 and Bax.
