中文摘要: 目的 分离、鉴定小花清风藤70%乙醇提取物的化学成分,并初步评价其体外抗氧化活性。方法采用硅胶、ODS反相硅胶、Sephadex-LH20柱色谱法及制备型高效液相色谱法等对小花清风藤70%乙醇提取物中化学成分进行分离和纯化,并结合核磁共振氢谱(1H-NMR)、核磁共振碳谱(13C-NMR)及电喷雾-质谱(ESI-MS)等波谱方法鉴定所分离出的化合物结构。分别采用2,2-联苯基-1-苦基肼基自由基(DPPH·)、2,2′-联氮基双(3-乙基苯并噻唑啉-6-磺酸)二铵盐自由基(ABST+)和羟基自由基(OH·)清除法考察各化合物的体外抗氧化活性。结果从小花清风藤70%乙醇提取物中共分离得到9个化合物,分别鉴定为芦丁(1)、邻苯二甲酸二异壬酯(2)、邻苯二甲酸二丁酯(3)、吐叶醇(4)、杜鹃醇(5)、槲皮素-3-O-龙胆双糖苷(6)、水仙苷(7)、山柰酚-3-O-芸香糖苷(8)、bonaroside(9)。体外抗氧化结果显示,化合物1~9均表现出一定的体外抗氧化活性,其中化合物1、6、7、8对DPPH·、ABST+、OH·的半数清除浓度均低于70μg/mL。结论首次从清风藤属植物中分离得到吐叶醇、杜鹃醇、bonaroside;芦丁、槲皮素-3-O-龙胆双糖苷、水仙苷、山柰酚-3-O-芸香糖苷表现出较好的体外抗氧化活性。
英文摘要: OBJECTIVE To separate and identif y the chemical constituen ts in 70% ethanol extract of Sabia parviflora ,and to preliminarily evaluate their in vitro antioxidant activity. METHODS The chemical constituents were separated and purified by silica gel,ODS reversed-phase silica gel ,Sephadex-LH20 column and preparative high performance liquid chromatography. The structures of the isolated compounds were identified by 1H-NMR,13C-NMR and ESI-MS. The in vitro antioxidant activities of the compounds were investigated by 2,2-diphenyl-1-picrylhydrazyl radical (DPPH·),2,2′-azino-bis(3-ethylbenzothiazoline-6- sulfonate)diammonium radical (ABST+)and hydroxyl radical (OH·). RESULTS A total of 9 compounds were isolated from the 70% ethanol extracts of S. parviflora . They were identified as rutin (1),diiononyl phthalate (2),dibutyl phthalate (3),vomifoliol (4),rhododendrol(5),quercetin-3-O-gentiobioside(6),narcissoside(7),kaempferol-3-O-rutinoside(8)and bonaroside (9). The in vitro antioxidant results showed that compound 1-9 showed certain in vitro antioxidant activity ,and the half scavenging concentrations of compound 1,6,7 and 8 to DPPH ·,ABST+,OH·were lower than 70 μg/mL. CONCLUSIONS Vomifoliol, rhododendrol and bonaroside are isolated from S. parviflora for the first time ,and rutin ,quercetin-3-O-gentiobioside,narcissoside and kaempferol- 3-O-rutinoside show good in vitro antioxidant activity.
