中文摘要: 目的:建立分析金钱草颗粒化学成分的方法。方法:采用高效液相色谱-电喷雾-飞行时间质谱(HPLC-ESI-Q-TOF-MS)法。色谱条件:色谱柱为Inertsil ODS-4,流动相为0.1%甲酸-甲醇(梯度洗脱),流速为0.8 ml/min,柱温为 30 ℃,进样量为15 μl;质谱条件:离子源为电喷雾离子原(负离子模式),端板偏移电压为-500 V,毛细管电泳电压为3 500 V,载气为氦气,雾化和干燥气体为高纯度氮气,流速为6 L/min,压力为1.0 bar,干燥气温度为180 ℃,扫描范围为50~1 500 m/z。采用ChemBioDraw Ultra 13.0和Bruker数据分析软件4.0版分析化学成分分子式。结果:从金钱草颗粒中鉴定出27种化学成分,分别为蔗糖、尿苷、没食子酸、新绿原酸、表没食子儿茶素、原儿茶酸、绿原酸、夏佛塔苷、咖啡酸、异夏佛塔苷、香草酸、阿魏酸、金丝桃苷、迷迭香酸、芦丁、异槲皮苷、紫云英苷、槲皮苷、杨梅素、对羟基苯甲酸、槲皮素、柚皮素、木犀草素、山柰酚、异鼠李素、芹菜素、大黄素。结论:咖啡酸、迷迭香酸、香草酸、表没食子儿茶素、尿苷、大黄素均为首次从金钱草制剂中被发现并报道。
英文摘要: OBJECTIVE:To establish a method for the analysis of chemical compositions in Jinqiancao granule. METHODS: HPLC-ESI-Q-TOF-MS was adopted. The chromatographic conditions: the column was Inertsil ODS-4 with mobile phase of 0.1% formic acid- methanol (gradient elution) at a flow rate of 0.8 ml/min, column temperature was 30 ℃, and the injection volume was 15 μl. MS conditions: ion source was ESI (negative ion mode), endplate offset voltage was -500 V, capillary electrophoresis was 3 500 V, carrier gas was helium gas, atomization and drying gas was high purity nitrogen gas at a flow rate of 6 L/min and a pressure of 1.0 bar, drying air temperature was 180 ℃. Scanning range was 50-1 500 m/z. ChemBioDraw Ultra13.0 and Bruker data analysis software 4.0 were used to analyze the chemical composition molecular formula. RESULTS: A total of 27 kinds of chemical components were identified in Jinqiancao granule, involving sucrose, uridine, gallic acid, new chlorogenic acid, (-)-epigallocatechin, protocatechuic acid, chlorogenic acid, schaftoside, caffeic acid, schaftoside, vanillc acid, ferulic acid, hyperin, rosmarinic acid, rutin, isoquercitrin, astragalin, quercetin-3-rhamnoside, myricetin, 4-hydroxybenzoic acid, quercetin, naringenin, luteolin, kaempferol, isorhmnetin, apigenin and emodin. CONCLUSIONS: Caffeic acid, rosmarinic acid, vanillic acid, (-)-epigallocatechin, uridine and emodin are firstly found and reported in the chemical compositions in Jinqiancao granule.
